Promising application of the SsCBF ELISA test to monitor the therapeutic response of feline sporotrichosis caused by Sporothrix brasiliensis from Brazilian epidemics

Sporotrichosis zoonotic transmission by cats has obtained hyperendemic magnitude in Rio de Janeiro, Brazil. Atypical cases, relapses, and reinfections as well as reduced diagnostic sensitivity of conventional methods have been reported. Previously, the anti-SsCBF enzyme-linked immunosorbent assay (ELISA) test was shown to be useful as a diagnostic tool for human sporotrichosis.
Effective diagnosis and treatment are critical to interrupt the chain of transmission of this major pathogen in Brazilian Public Health. To evaluate its applicability for feline sporotrichosis diagnosis and/or therapeutic follow-up, 15 domestic cats from Rio de Janeiro were clinically and laboratory monitored by cytopathology, culture, Sporothrix genotyping, and anti-SsCBF IgG levels.
Subsequently, animals were divided into satisfactory and non-satisfactory therapeutic responders. Averages of antibody serum levels obtained for diagnosis (first consultation) compared with the levels found after follow-up (last consultation) were significantly different in both groups (p = 0.0002 and p = 0.038, respectively). We conclude that the SsCBF ELISA test can predict feline sporotrichosis therapeutic responses even for animals with distinct clinical evolutions.

Repeatability of a Commercially Available ELISATest for Determining the Herd-Level Salmonella enterica subsp. enterica Serovar Dublin Status in Dairy Herds Using Bulk Milk

An Enzyme-Linked Immunosorbent Assay (ELISA) is currently available for detection of antibodies against Salmonella Dublin in bovine milk. However, when used in a surveillance program, samples may undergo various storage conditions.
The objective of this study was to estimate the repeatability of an ELISA test when used on fresh and frozen samples. Each of 845 bulk milk collected samples was subdivided into 3 aliquots and analyzed using PrioCHECK™ Salmonella Ab Bovine Dublin. ELISA percent positivity results (PP%) were compared between aliquots submitted to the initial analysis and a second analysis conducted 24 h later.
The third aliquots were either preserved for 13-14 days (n = 413) or 25-28 days (n = 432) at -20°C prior to analysis and results were compared to the initial analysis. There was excellent concordance between the two initial values and with values obtained after 13-14 and 25-28 days-freezing.
The corresponding concordance correlation coefficients were 0.96, 0.97, and 0.94, respectively. Bland-Altman plots showed differences of PP% of 0.1 percentage points on average between the initial and second fresh samples. Freezing for 13-14 and 25-28 days led to overestimation of the initial values by 0.1, and 0.4 percentage points, respectively.
Regarding the classification of samples, greater disagreement was observed between 25 and 28 days-frozen and initial samples when using the cut-off 15% (kappa = 0.76) compared to 35% (kappa = 0.90). Our study showed that PrioCHECK™ has good repeatability and that frozen bulk milk samples could generate reliable results. However, the larger variability at lower PP% should be considered when setting up a threshold.

Individual and herd-level milk ELISA test status for Johne’s disease in Ireland after correcting for non-disease-associated variables

Antibody-detecting tests for Mycobacterium avium ssp. paratuberculosis (MAP) have low sensitivity and imperfect specificity for detection of infection. Sensitivity increases as the disease progresses. Aside from infection status and stage of disease, several factors affect test performance.
These factors have not yet been studied in dairy cows producing lower volumes of milk with higher solids concentration, such as those managed in low-input, pasture-based production systems.
Furthermore, the effect of correcting for these associations on individual and herd test status is also unknown. The first objective of this study was to examine the relationship between MAP antibody response in milk and milk yield, somatic cell count (SCC), fat and protein contents, and stage of lactation in dairy cows enrolled in the national Johne’s Disease Control Programme (JDCP) in Ireland.
The second objective was to examine the effect of correcting the antibody response for these associations on the test status of individual cows and herds, given that individual tests are often used to define a herd’s status. Data were extracted for herds in the JDCP from January 2014 to December 2015 inclusive, consisting of 42,657 milk recordings from 18,569 cows across 187 dairy herds.
Two linear regression models were constructed to investigate the association between log-transformed MAP sample-to-positive ratio and milk recording data and in primi- and multiparous cows.
Days in milk was modeled as a B-spline in each model, and cow and herd were included as random effects. Across both models, natural log-transformed MAP antibody response was negatively associated with milk yield, positively associated with protein and fat production, and had a curvilinear association with log-transformed SCC.
The association between MAP antibody response and days in milk varied over the course of the lactation. However, when combined, these variables explained only 5.1% of the variation in the antibody response of the population. After correcting for these associations, 93 multiparous cows and 20 primiparous cows changed category (negative, suspect, or positive).
When considered at the herd-test level, out of a total of 531 herd tests, 1 herd changed from negative to positive, and 5 herds changed from positive to negative. This study provides useful information to aid in the interpretation of antibody results for herds testing animals for the presence of MAP infection. At an overall population level, correction of the serological response for non-disease-associated factors has the potential to change the status of only a small number of cows.
At the herd level, the proportion of herds changing status was minimal. However, depending on the implications of a herd-level serological diagnosis, consideration should be given to correcting for these non-disease-associated variables within the context of national JD control programs.
